Just to be sure, since the vision analysis is done using grayscale, is it true that it doesn't matter if the image trackable is printed in color or grayscale?

This is a good thing to be aware of, that the image will be transformed to grayscale, because it will help in evaluating whether images will make suitable targets. The Tracker is looking for variations in luminance contrast, which is why grayscale is used. And by accentuating the contrast of your image, or areas of your image, you can often improve its tracking performance.

I've tested with printed out greyscale image ,while my trackable is done with a 16bit colour image. It does work!

Yes, it doesn't matter. The color image will be transformed to grayscale by the SDK.
